Resumen:
Advanced Microscopy Facility arises from the need to centralize highly complex microscopy equipment and maximize use, management, maintenance and growth potential. The instruments of this facility are enrolled in the National Microscopy System, network promoted by the CONICET and MinCyT. The creation of the Facility was achieved by the combination of two multi - institutional consortia (PME2006 and PAE- PME 2007 grants) coordinated by professors of the Faculty of Exact Sciences (UNLP). General objectives are to bring close to the science community and guarantee the proper use of highly complex equipment, give training courses and interact with others facilities. The main instruments are:?Leica TCS SP5 Confocal Spectral MicroscopeConfocal microscopy has advantages over conventional optical microscopy since the use of a pinhole excludes unfocused light resulting in an increase in axial resolution of the image. The Leica TCS SP5 confocal microscope covers a wide range of imaging requirements with excellent performance, the system provides a full range of scanning speeds even at the highest resolution. It allows three-dimensional, spectral and time acquisitions. It is equipped with 8 lines of laser excitation (458-475 - 488-496 - 514-543 - 594-633 nm), three simultaneous detection channels; also has an UV lamp for epifluorescence visualization. It has three objectives (10X DRY - NA: 0.30, 20X WATER - NA: 0.70, 63X OIL - NA: 1.4) and a motorized stage.?Leica Laser Disection Microscope This is a contact- and contamination-free method for isolating specific single cells or entire areas of tissue from a wide variety of tissue samples. Isolated areas are suitable for molecular analysis in any of the following fields: Molecular Pathology, Cell Biology, Cancer Research, Forensic Medicine, Genomics / Proteomics, Immunology, Food Research, Neuroscience , prenatal diagnosis, Microbiology and Virology. The system provides high image quality for locating cells quickly and easily. The specimens are mounted on a membrane, drawing freehand or by default figures can select as many individual cells or groups of cells as desired. The selection is automatically cut using a solid high-precision UV laser. ?Thermo-Shandon CryotomeThis equipment is designed to cut frozen specimens rapidly and accurately so that later examined under a microscope. Processing histological preparations obtained by freezing can complement the ability to perform immunofluorescent histochemistry techniques and nucleic acid recovery after dissection microscopy. ?Nikon Ti Eclipse Inverted Fluorescence MicroscopeThis microscope allows the observation of both samples on slides as in petri dishes or well plates. Allows the observation of live cells immersed in medium. Since it has a camera model IT -U Eclipse NIKON is possible to acquire images from the observed samples. It has an epi-fluorescence system and phase contrast.
