In yeast actively growing cells, Maf1 localizes into the cytoplasm as a phosphorilated protein. Under unfavorable growing conditions it becomes dephosphorilated and is subsequently transported to the nucleus, where it represses transcription. Maf1 is widely distributed along eukaryotic organisms, although it has been only studied in human and yeast models. We identified a putative orthologue in T. cruzi genome (TcMaf1). TcMaf1 could not complement S. cerevisiae ΔMaf mutant, probably due to a high sequence divergence. Cytolocalization experiments with polyclonal antibodies raised against the recombinant protein showed that TcMaf1 displays cytoplasmic localization in metacyclic trypomastigotes, amastigotes and epimastigotes. Cl-Brenner pLEW epimastigotes were transfected with the tetracycline inducible pINDEX-I vector, containing TcMaf1 and TcMaf1-cmyc. Induced parasites had identical growth rates compared to those not induced. Confocal microscopy showed TcMaf1-myc is located all over the parasite body in discrete foci, excluded from the nucleus and the mitochondrion. Under different stress conditions, we were not able to demonstrate that this protein could be located into the nucleus at some time point. Also, yeast two-hybrid assays with putative T. cruzi ARNP III transcription basal factors failed to detect protein-protein interactions with TcMaf1, neither it interacted with TcPuf6 or TcDhh1, two proteins associated with cytoplasmic mRNAs. Which is the role of TcMaf1 in trypanosomes remains an open question. Presently we are characterizing Maf1 RNAi knockdown procyclic T. brucei. Co-localization studies with known cytoplasmic proteins are also under schedule.
