Resumen:
Dendritic cells (DC) are the most potent Ag presenting cells.We have previously demonstrated that vaccination with DC charged with apoptotic/necrotic (Apo/Nec) melanoma B16/F1 cells provides long term protection against melanoma, and that a tertiary lymphoid structure is formed at the vaccination site. However, when DC are cocultured with Apo/Nec cells, their stimulating capacity diminishes 3.7 fold (p0.05). To increase DC stimulating capacity, cocultures in the presence of 10μg/ml imiquimod (Imi) (toll-like receptor 7 ligand), 10μg/ml poly I:C (toll-like receptor 3 ligand), 30μg/ml prostaglandin E2 (PGE2), or a mixture of the three were performed.
